Abstract:
In Oesterreich sind etwa 100.000 Personen von DAT (Demenz vom Alzheimer-Typ) betroffen. Pathologisches Hauptmerkmal sind extrazellulaere Gehirnablagerungen, die vorwiegend aus dem Protein #beta#-Amyloid bestehen. Neueste Ergebnisse unterstuetzen die Hypothese, dass aggregiertes #beta#-Amyloid eine entscheidende Rolle in der Entstehung und dem Verlauf der Krankheit spielt. Im Rahmen dieser Arbeit untersuchten wir den Einfluss der Isoform #beta#-Amyloid (1-40) auf die Integritaet der Plasmamembran neuronaler Zellen, am Beispiel der Zelllinie SH-SY5Y. Zugabe von aggregiertem #beta#-Amyloid (1-40) bewirkte eine Erniedrigung der Membranfluiditaet, eine Reduzierung der Membranimpermeabilitaet, sowie einen Anstieg oxidierter Membranlipide. Letztere Beobachtung ist von besonderem Interesse, da #beta#-Amyloid (1-40) unabhaengig des Aggregationszustandes eine Abnahme intrazellulaerer Peroxyde in ruhenden und H_2O_2-behandelten SH-SY5Y Zellen induzierte und daher antioxidative Eigenschaften zeigte. Weiters beobachteten wir quantitative Aufnahme und Abbau von #beta#-Amyloid (1-40), sowie die Freisetzung der Isoform #beta#-Amyloid (1-42) durch H_2O_2-Stimulation in SH-SY5Y Zellen und Vollblut. Diese Ergebnisse bekraeftigen die Annahme, dass Neuronen eine aktive Rolle im #beta#-Amyloid-Metabolismus spielen. Schlussendlich fuehren unsere gewonnenen Daten zu folgendem Modell: Um gegen den erhoehten oxidativen Stress im Alter anzukaempfen, setzen Neuronen (und andere im Blut vorhandene Zellen) antioxidatives, monomeres #beta#-Amyloid frei. Bei fortdauerndem oxidativen Einfluss aggregiert akkumuliertes #beta#-Amyloid und erlangt dadurch prooxidative und neurotoxische Eigenschaften.

Alzheimer disease is a devastating neurodementia. Pathological hallmark are extracellular plaques in the brain, mostly constituted of the peptide #beta#-Amyloid. Recent data supports the hypothesis that aggregated #beta#-Amyloid is neurotoxic. In this study, we focused on the impact of #beta#-Amyloid (1-40) on the integrity of the plasma membrane in the neuronal cell line SH-SY5Y. Addition of aggregated #beta#-Amyloid (1-40) altered the membrane fluidity, induced leakage of membrane constituents, and oxidized membrane lipids. Interestingly, the monomeric peptide also decreased the concentration of intracellular reactive oxygen species in untreated and H_2O_2-treated cells, thereby acting as anti-oxidant. Moreover, we characterized the metabolism of #beta#-Amyloid in SH-SY5 cells. The cells degraded a significant amount of #beta#-Amyloid (1-40) and released measurable amounts of the (1-42) isoform into the extra-cellular fluid upon addition of H_2O_2, thereby contributing actively to the increased burden of #beta#-Amyloid in the Alzheimer brain. Finally our observation that ex vivo addition of H_2O_2 to whole blood increased the concentration of plasma #beta#-Amyloid leads us to the following model: Anti-oxidative, monomeric #beta#-Amyloid is released to fight against oxidative stress that occurs in the elderly. Ultimately, accumulated #beta#-Amyloid aggregates and becomes neurotoxic by acting as a pro-oxidant and damaging the neuronal membrane.
