In der vorliegenden Arbeit wurde für die quantitative Analyse von pharmazeutischen und medizinischen zellbiologischen Fragestellungen ein Impedanzanalysesystem mit einem Multisensor-Interface entwickelt.<br />Die Impedanzspektroskopie eignet sich hervorragenden, um nicht invasive in-vitro-Untersuchungen an lebenden Zellkulturen durchzuführen. Zur Charakterisierung des Impedanzanalysesystems wurde die CaCo2-Zelllinie verwendet, welche die Eigenschaften des Dünndarmepithelgewebes widerspiegelt. Damit wird es möglich humanspezifische Fragestellungen wie z. B. die Resorptionsfähigkeit oder die Regenerationsfähigkeit des Darmtraktes, nicht nur qualitativ sondern auch quantitativ zu analysieren.<br />Mit dem zum Einsatz kommenden impedanzspektroskopischen Verfahren ist nicht nur eine kontinuierliche Langzeitmessung sondern auch eine präzis auflösende Analyse möglich. Hierfür kommen mikrotechnologisch strukturierte planare Interdigitated Electrodes (IDES) zum Einsatz. Für die Langzeitmessungen wurden zwei Elektrodenpaardesigns eingesetzt, welche jeweils vier getrennte Zellkulturen auf einem Multi Electrode Array, kurz MEA-Chip, vereinen. Die Anschlussgeometrie des MEA-Chips basiert auf dem MEA60 Biochipstandard von Multichannel Systems. Diese beiden Designs wurden hinsichtlich ihrer Sensitivität und Auflösung miteinander verglichen. In dem IDES-4-Elektrodenpaardesign des einen MEA-Chips ist für jede Zellkultur ein Elektrodenpaar vorgesehen. Beim IDES-8-Elektrodenpaardesign des anderen MEA-Chipdesigns wurden pro Zellkultur je zwei Elektrodenpaare implementiert. Durch diese Weiterentwicklung gelang es zum einen eine höhere Auflösung zu erreichen und zum anderen räumliche Unterschiede in derselben Zellkultur detektieren zu können. Durch die höhere Auflösung sind zellwachstums- und umgebungsbedingte biologische Schwankungen besser erkennbar.<br />Die Versorgung von lebenden Zellen mittels Nährstoffen ist für Langzeitmessungen von essentieller Bedeutung. Das vorliegende Impedanzanalysesystem, welches über eine PC-Anbindung eine online-Analyse erlaubt, wurde deshalb um ein mikrofluidisches Versorgungssystem erweitert. Das Versorgungssystem besteht aus einer PC-gesteuerten hochpräzisen Spritzenpumpe und einem geschlossenen mikrofluidischen Zellkulturaufsatz. Mit der mikrofluidischen Versorgung kann zum einen eine Mediumsversorgung der Zellen über den Kultivierungszeitraum erfolgen und zum anderen können die vier Einzelreservoirs des MEA-Chips manuell und getrennt voneinander angesteuert werden. Damit die neuartige Medienversorgung mit dem Impedanzanalysesystem gleichzeitig gesteuert werden kann, wurde eine zusätzliche Softwaresteuerung implementiert. Demzufolge kann das komplette Impedanzanalysesystem über einen PC gesteuert und die Messdaten ausgewertet werden.<br />Die durchgeführten Messungen und Analysen des CaCo2-Zelllayers zeigen den Vorteil des besser aufgelösten IDES-8-Elektrodenpaardesigns und die Funktionalität des mikrofluidischen Versorgungssystems. Des Weiteren wurden erfolgreiche Permeabilitätsmessungen mit Bromelain als Permeation Enhancer und Regenerationsfähigkeitsmessungen nach mechanischen Beschädigungen der Zellschicht analysiert. Für die Grundlagenforschung ist das Wissen über den Einfluss von pharmazeutischen Präparaten auf die Permeabilität des Darmtrakts und die Regenerationsfähigkeit der Darmwand nach mechanischen Beschädigungen für neuartige Heilungsmethoden und medikamentöse Behandlungen besonders interessant. Mit den vorliegenden Messungen kann die Besonderheit der Impedanzspektroskopie, welche quantitativ messbare Ergebnisse liefert, gezeigt werden.<br />

In this thesis an impedance-analyzer-system was developed and used for long-time impedance measurements of living epithelial tissue.<br />The measurement of impedance spectra of cell cultures allows to address a wide range of pharmaceutical and medical cytological issues. As (f. e.<br />on living cell cultures) most conventional analyses are based on qualitative optical results impedance spectroscopy offers an excellent instrument for non-invasive in-vitro investigations. The cells used for this study are the CaCo2-cellline. This cell culture has well defined metabolic and cell-cell contact properties comparable to the human small intestine epithelial cell walls. Combining the CaCo2-cells with the measurement system mentioned above allows to investigating important biological problems such as drug intake and regeneration properties.<br />The use of microstructural and planar interdigitated electrodes (IDES) provides low signal-to-noise ratio and allows long-time measurement. The IDES based on gold and are fabricated on glass-substrates with microstructural techniques. In the thesis two different electrode designs based on four separated culture wells with 8mm in diameter are presented. The wells are independent from each other and constitute one single cell culture. Using the 4-electrodepair-design each cell culture can be investigated by one electrode only. In contrast to this procedure the further developed 8-electrodepair-design provides two high resolving electrodes for each cell culture. This offers a better spatial resolution of culture-specific proliferation variations and a higher cell-cell contact resolution. This enhances the possibility of detecting environmental changes within the cell culture and provides for a better monitoring of intestinal drug intake. The two electrode designs are compared in order to evaluate different functionalities and resolutions.<br />The contact pad is adapted to the Multichannel System MEA60 biochip standard.<br />The impedance-analyzer-system contains a specific software solution for monitoring PC-based long-time measurement. By implementing a microfluidic media supply made of biocompatible materials the system can be extended. The microfluidic system includes a high-precision syringe pump and an extended cell-sensor-design. With this system a continuous media supply over the whole culture period is possible. The flow into separated culture wells can be selected manually, thus simulating different cell environments. For the whole system the new microfluidic device includes a control-software to guarantee a continuous online-monitoring. Therefore it is possible to analyze the measured data in real-time. For a successfully measurement system it is necessary to take care of biocompatible materials in the whole system. With this requirement all components such as gold-electrodes, glass-substrate, the PDMS-cell-culture-system, microfluidic components such as silicon tubes and steel-alloys hollow needles are biocompatible.<br />Finally it can be stated that the measurements carried out with the 8-electrode-design have the advantage it can be concluded of higher resolution. The functionality of the microfluidic supplying device and its control-software has been demonstrated. Furthermore, successful permeability measurements with Bromelain as permeation enhancer as well as measurements of the regenerative properties of the CaCo2-cellmonolayer could be demonstrated. With the improved impedance-analyzer-system not only a good quantitative diagnostic instrument has been implemented but it is also provided for a system which enables reproducible results in the biological distribution.<br />
