Biokompatible Membranen mit definierten Porengrößen sind für zahlreiche biomedizinische Applikationen eine Grundvoraussetzung. Die Herstellung und Handhabung solcher Membranen mit exakt definierter Porengröße ist eine technologische Herausforderung, sowohl in mechanischer als auch chemischer, biologischer und fertigungstechnischer Hinsicht. In dieser Arbeit wird ein Zellkulturchip, bestehend aus einem Nervenisolationsaufsatz (NI-Device) und einem Mikroelektroden Array (MEA), durch einen Versorgungskanal erweitert, welcher durch eine solche Mikroporöse Membran von den Mikrokanälen des NI-Device getrennt ist. Der Schwerpunkt liegt hierbei auf der Konzeption der Membranfertigung, Modellierung des Diffusionsstoffstroms und der Integration dieser Membran in das bestehendes NI-Device. Hierfür wird eine mikroporöse Membran aus biokompatiblen PDMS entwickelt und es wird gezeigt, wie die Membran in das NI-Device integriert wird, unter der Beibehaltung des ursprünglichen NI-Designs. Dabei werden verschiedene Methoden zur Mikrostrukturierung und Oberflächenmodifikation, sowie messtechnischen Untersuchung vorgestellt und angewendet. Die Entwicklung wird durch Multiphysik-Simulationen unterstützt und die praktische Umsetzung durch einen Farbmitteltest validiert. Die Prozessschritte zur Fertigung der einzelnen Komponenten, sowie zur Fertigung des gesamten Systems wurden zu detaillierten Anleitungen zusammengefasst und ermöglichen einen Nachbau für nachfolgende Arbeiten oder Erweiterungen.

Biocompatible membranes with defined pore sizes are a basic requirement for numerous biomedical applications. The fabrication and the handling of such membranes with defined pore size is technologically challenging in respect to mechanical, chemical and biological aspects, as well as in respect to manufacturing regards. In this work a cell culture chip, composed of a nerve isolation device (NI-Device) and Microelectrode Array (MEA), is extended with an additional supply channel that is separated from the microchannels of the NI-Device by a microporous membrane. This work will focus on the process development for the fabrication of a porous membrane, modeling of the diffusion flux and integration of the new components into the existent NI-Device. For this purpose, a biocompatible microporous polydimethylsiloxane (PDMS) membrane will be developed and it will be shown how to integrate it in the existent NI-Device with retaining the NI-Devices basic design. Thereby different methods for microstructuring and surface modification will be presented and implemented, as well as different measurement technologies. The development will be supported by multiphysics simulations and the practical realization of the whole system will be validated by a pigment solution test. The process steps for the fabrication of the particular components, as well as the complete system are summarized to detailed recipes. This should allow a rebuild for following works or extensions.
